Margaret Smith (AUS) defeated Maria Bueno (BRA) 6–4, 7–5 in the final to win the Ladies' Singles tennis title at the 1965 Wimbledon Championships.

Van Zyl, who withdrew because of a family bereavement, was replaced by a Lucky Loser Michelle Boulle (CHI). For the first time in the history of the All England Championships at Wimbledon, there was no British player seeded in the Ladies Singles Championship. Despite this, ironically a British player progressed to the last four unseeded.
